INDICATIONS AND USAGE<\/strong><\/strong><\/span><\/p>\n

LuciBru is a kinase inhibitor indicated for the treatment of:<\/span><\/p>\n

\u2022 Adult patients with mantle cell lymphoma (MCL) who have received at least one prior therapy .<\/span><\/p>\n

This indication is approved under accelerated approval based on overall response rate. Continued approval for this indication may be contingent upon verification and description of clinical benefit in a confirmatory trial(s).<\/span><\/p>\n

\u2022 Adult patients with chronic lymphocytic leukemia (CLL)\/Small lymphocytic lymphoma (SLL) .<\/span><\/p>\n

\u2022 Adult patients with chronic lymphocytic leukemia (CLL)\/Small lymphocytic lymphoma (SLL) with 17p deletion.<\/span><\/p>\n

\u2022 Adult patients with Waldenstr\u00f6m\u2019s macroglobulinemia (WM) .<\/span><\/p>\n

\u2022 Adult patients with marginal zone lymphoma (MZL) who require systemic therapy and have received at least one prior anti-CD20-based therapy .<\/span><\/p>\n

This indication is approved under accelerated approval based on overall response rate. Continued approval for this indication may be contingent upon verification and description of clinical benefit in a confirmatory trial(s).<\/span><\/p>\n

\u2022 Adult and pediatric patients age 1 year and older with chronic graft versus host disease (cGVHD) after failure of one or more lines of systemic therapy .<\/span><\/p>\n

 <\/span><\/p>\n

DOSAGE AND ADMINISTRATION <\/strong><\/strong><\/span><\/p>\n

\u2022 MCL<\/span> <\/span><\/span>and<\/span> <\/span><\/span>MZL<\/span>: <\/span>560 <\/span>mg <\/span>taken <\/span>orally <\/span>once <\/span>daily.<\/span><\/span><\/p>\n

\u2022 CLL\/SLL<\/span> <\/span><\/span>and<\/span> <\/span><\/span>WM<\/span>: <\/span>420 <\/span>mg <\/span>taken <\/span>orally <\/span>once <\/span>daily.<\/span><\/span><\/p>\n

\u2022 <\/span>cGVHD<\/span><\/span>:<\/span><\/span><\/p>\n

o Patients <\/span>12 <\/span>years <\/span>and <\/span>older: <\/span>420 <\/span>mg taken <\/span>orally <\/span>once <\/span>daily.<\/span><\/span><\/p>\n

o Patients <\/span>1 <\/span>to <\/span>less <\/span>than <\/span>12 <\/span>years <\/span>of <\/span>age: <\/span>240 <\/span>mg\/m2<\/span><\/sup> <\/span>taken <\/span>orally <\/span>once <\/span>daily (up to a dose of 420 mg).<\/span><\/p>\n

Tablets <\/span>should <\/span>be <\/span>taken <\/span>orally <\/span>with <\/span>a <\/span>glass <\/span>of <\/span>water. <\/span>Do not cut, crush, or chew the tablets. <\/span><\/p>\n

WARNINGS AND PRECAUTIONS <\/strong><\/strong><\/span><\/p>\n

\u2022 Hemorrhage<\/span>: <\/span>Monitor <\/span>for <\/span>bleeding <\/span>and <\/span>manage.<\/span><\/span><\/p>\n

\u2022 Infections<\/span>: <\/span>Monitor <\/span>patients <\/span>for <\/span>fever <\/span>and <\/span>infections, <\/span>evaluate <\/span>promptly, <\/span>and treat.<\/span><\/p>\n

\u2022 Cardiac<\/span> <\/span><\/span>Arrhythmias,<\/span> <\/span><\/span>Cardiac<\/span> <\/span><\/span>Failure,<\/span> <\/span><\/span>and<\/span> <\/span><\/span>Sudden<\/span> <\/span><\/span>Death<\/span>: <\/span>Monitor <\/span>for <\/span>symptoms of arrhythmias and cardiac failure and manage.<\/span><\/p>\n

\u2022 Hypertension<\/span>: <\/span>Monitor <\/span>blood <\/span>pressure <\/span>and <\/span>treat.<\/span><\/span><\/p>\n

\u2022 Cytopenias<\/span>: <\/span>Check <\/span>complete <\/span>blood <\/span>counts <\/span>monthly.<\/span><\/span><\/p>\n

\u2022 Second<\/span> <\/span><\/span>Primary<\/span> <\/span><\/span>Malignancies<\/span>: <\/span>Other <\/span>malignancies <\/span>have <\/span>occurred <\/span>in <\/span>patients, including skin cancers, and other carcinomas.<\/span><\/p>\n

\u2022 Tumor<\/span> <\/span><\/span>Lysis<\/span> <\/span><\/span>Syndrome<\/span> <\/span><\/span>(TLS)<\/span>: <\/span>Assess <\/span>baseline <\/span>risk <\/span>and <\/span>take <\/span>precautions. <\/span>Monitor and treat for TLS.<\/span><\/p>\n

\u2022 Embryo-Fetal Toxicity<\/span>: Can cause fetal harm. Advise females of <\/span>reproductive <\/span>potential <\/span>of <\/span>the <\/span>potential <\/span>risk <\/span>to <\/span>a <\/span>fetus <\/span>and <\/span>to <\/span>use <\/span>effective <\/span>contraception.<\/span><\/p>\n

 <\/span><\/p>\n

ADVERSE REACTIONS <\/strong><\/strong><\/span><\/p>\n

\u2022 The most common (\u226530%) adverse reactions in patients with B-cell <\/span>malignancies <\/span>(MCL, <\/span>CLL\/SLL, <\/span>WM <\/span>and <\/span>MZL) <\/span>are <\/span>thrombocytopenia, <\/span>diarrhea, fatigue, musculoskeletal <\/span>pain, <\/span>neutropenia, rash, anemia, and <\/span>bruising.<\/span><\/p>\n

\u2022 The <\/span>most <\/span>common <\/span>(\u226520%) <\/span>adverse <\/span>reactions <\/span>in <\/span>adult <\/span>or <\/span>pediatric <\/span>patients <\/span>with cGVHD are fatigue, anemia, bruising, diarrhea, thrombocytopenia, <\/span>musculoskeletal pain, pyrexia, muscle spasms, stomatitis, hemorrhage, <\/span>nausea, abdominal pain, pneumonia, and headache.<\/span><\/p>\n

DRUG INTERACTIONS <\/strong><\/strong><\/span><\/p>\n

\u2022 CYP3A <\/span>Inhibitors: <\/span>Modify <\/span>LuciBru <\/span>dose <\/span>as <\/span>described.<\/span><\/span><\/p>\n

\u2022 CYP3A <\/span>Inducers: <\/span>Avoid <\/span>coadministration <\/span>with <\/span>strong <\/span>CYP3A <\/span>inducers.<\/span><\/span><\/p>\n

 <\/span><\/p>\n

USE IN SPECIFIC POPULATIONS <\/strong><\/strong><\/span><\/p>\n

\u2022 Lactation<\/span>: <\/span>Advise <\/span>not <\/span>to <\/span>breastfeed. <\/span><\/span><\/p>\n

\u2022 Hepatic Impairment<\/span>: Avoid use of LuciBru in patients with severe <\/span>hepatic <\/span>impairment. <\/span>In <\/span>patients <\/span>with <\/span>mild <\/span>or <\/span>moderate <\/span>impairment, <\/span>reduce <\/span>LuciBru dose.<\/span><\/p>\n
