INDICATIONS AND USAGE<\/span><\/strong><\/strong><\/span><\/p>\n

LuciRip is a kinase inhibitor indicated for the treatment of adult patients with advanced gastrointestinal stromal tumor (GIST) who have received prior treatment with 3 or more kinase inhibitors, including imatinib.<\/span><\/p>\n

DOSAGE AND ADMINISTRATION<\/span><\/strong><\/strong><\/span><\/p>\n

Recommended Dosage: 150 mg orally once daily with or without food.<\/span> <\/span><\/span><\/p>\n

WARNINGS AND PRECAUTIONS<\/span><\/strong><\/strong><\/span><\/p>\n

\u00b7<\/span> Palmar-Plantar Erythrodysesthesia Syndrome: Based on severity, withhold LuciRip and resume at same or reduced dose.<\/span><\/span><\/p>\n

\u00b7<\/span> New Primary Cutaneous Malignancies: Perform dermatologic evaluations when initiating LuciRip and routinely during treatment.<\/span><\/span><\/p>\n

\u00b7<\/span> Hypertension: Do not initiate LuciRip in patients with uncontrolled hypertension and monitor blood pressure during treatment. Based on severity, withhold LuciRip and then resume at same or reduced dose or permanently discontinue.<\/span><\/span><\/p>\n

\u00b7<\/span> Cardiac Dysfunction: Assess ejection fraction by echocardiogram or MUGA scan prior to initiating LuciRip and during treatment, as clinically indicated. Permanently discontinue LuciRip for Grade 3 or 4 left ventricular systolic dysfunction.<\/span><\/span><\/p>\n

\u00b7<\/span> Risk of Impaired Wound Healing: Withhold LuciRip for at least 1 week prior to elective surgery. Do not administer for at least 2 weeks after major surgery and until adequate wound healing. The safety of resumption of LuciRip after resolution of wound healing complications has not been established.<\/span><\/span><\/p>\n

\u00b7<\/span> Photosensitivity: May cause photosensitivity reactions. Advise patients to limit direct ultraviolet exposure.<\/span><\/span><\/p>\n

\u00b7<\/span> Embryo-Fetal Toxicity: Can cause fetal harm. Advise females of reproductive potential of the potential risk to a fetus and to use effective contraception.<\/span><\/span><\/p>\n

 <\/span><\/p>\n

ADVERSE REACTIONS<\/span><\/strong><\/strong><\/span><\/p>\n

The most common adverse reactions (\u226520%) were alopecia, fatigue, nausea, abdominal pain, constipation, myalgia, diarrhea, decreased appetite, palmarplantar erythrodysesthesia, and vomiting. The most common Grade 3 or 4 laboratory abnormalities (\u22654%) were increased lipase and decreased phosphate.<\/span><\/p>\n

DRUG INTERACTIONS<\/span><\/strong><\/strong><\/span><\/p>\n

\u00b7<\/span> Strong CYP3A Inhibitors: Monitor more frequently for adverse reactions.<\/span><\/span><\/p>\n

\u00b7<\/span> Strong CYP3A Inducers: Avoid concomitant use of strong CYP3A inducers.<\/span><\/span><\/p>\n

\u00b7<\/span> Moderate CYP3A Inducers: Avoid concomitant use of moderate CYP3A inducers. If a moderate CYP inducer cannot be avoided, increase ripretinib dose frequency to twice daily.<\/span><\/span><\/p>\n

USE IN SPECIFIC POPULATIONS<\/span><\/strong><\/strong><\/span><\/p>\n

Lactation: Advise not to breastfeed.<\/span><\/p>\n

Storage<\/span><\/strong><\/strong><\/span><\/p>\n

Store at 20<\/span>\u2103<\/span> to 25<\/span>\u2103<\/span> (68<\/span>\u2109<\/span> to 77<\/span>\u2109<\/span>), excursions permitted between 15<\/span>\u2103<\/span> and 30<\/span>\u2103<\/span> (59<\/span>\u2109<\/span> and 86<\/span>\u2109<\/span>) [see USP Controlled Room Temperature]. Protect from moisture. <\/span><\/span><\/p>\n
