 <\/span><\/p>\n

INDICATIONS AND USAGE<\/strong><\/strong><\/span><\/p>\n

LuciAva is a kinase inhibitor indicated for:<\/span><\/p>\n

Gastrointestinal Stromal Tumor (GIST)<\/span><\/p>\n

\u2022 the treatment of adults with unresectable or metastatic GIST harboring a platelet-derived growth factor receptor alpha (PDGFRA) exon 18 mutation, including PDGFRA D842V mutations. <\/span><\/p>\n

Advanced Systemic Mastocytosis (AdvSM).<\/span><\/p>\n

\u2022 the treatment of adult patients with AdvSM. AdvSM includes patients with aggressive systemic mastocytosis (ASM), systemic mastocytosis with an associated hematological neoplasm (SMAHN), and mast cell leukemia (MCL). <\/span><\/p>\n

\u2022 Limitations of Use: LuciAva is not recommended for the treatment of patients with AdvSM with platelet counts of less than 50\u00d7109<\/span><\/sup>\/L<\/span><\/p>\n

Indolent Systemic Mastocytosis (ISM)<\/span><\/p>\n

\u2022 the treatment of adult patients with ISM.<\/span><\/p>\n

\u2022 Limitations of Use: LuciAva is not recommended for the treatment of patients with ISM with platelet counts of less than 50\u00d7109<\/span><\/sup>\/L.<\/span><\/p>\n

 <\/span><\/p>\n

DOSAGE AND ADMINISTRATION<\/strong><\/strong><\/span><\/p>\n

\u2022 GIST: Select patients for treatment with LuciAva based on the presence of a PDGFRA exon 18 mutation. <\/span><\/p>\n

\u2022 GIST: The recommended dosage is 300 mg orally once daily.<\/span><\/p>\n

\u2022 AdvSM: The recommended dosage is 200 mg orally once daily.<\/span><\/p>\n

\u2022 ISM: The recommended dosage is 25 mg orally once daily. <\/span><\/p>\n

\u2022 Patients with severe hepatic impairment (Child-Pugh Class C): reduce dose of LuciAva.<\/span><\/p>\n

 <\/span><\/p>\n

DOSAGE FORMS AND STRENGTHS <\/strong><\/strong><\/span><\/p>\n

Tablets: 100 mg\u00d730 tablets<\/span><\/p>\n

 <\/span><\/p>\n

CONTRAINDICATIONS <\/strong><\/strong><\/span><\/p>\n

 <\/span><\/p>\n

WARNINGS AND PRECAUTIONS <\/strong><\/strong><\/span><\/p>\n

\u2022 Intracranial Hemorrhage: Permanently discontinue for any occurrence of any grade. <\/span><\/p>\n

\u2022 Cognitive Effects: A broad spectrum of cognitive adverse reactions can occur in patients receiving LuciAva. In patients with GIST, AdvSM, or ISM depending on the severity, continue LuciAva at same dose, withhold and then resume at same or reduced dose upon improvement, or permanently discontinue.<\/span><\/p>\n

\u2022 Photosensitivity: May cause photosensitivity reactions. Advise patients to limit direct ultraviolet exposure. <\/span><\/p>\n

\u2022 Embryo-Fetal Toxicity: Can cause fetal harm. Advise females and males of reproductive potential of the potential risk to a fetus and to use effective contraception.<\/span><\/p>\n

 <\/span><\/p>\n

ADVERSE REACTIONS <\/strong><\/strong><\/span><\/p>\n

The most common adverse reactions are: <\/span><\/p>\n

\u2022 GIST (\u226520% incidence): edema, nausea, fatigue\/asthenia, cognitive impairment, vomiting, decreased appetite, diarrhea, increased lacrimation, abdominal pain, constipation, rash, dizziness, and hair color changes. <\/span><\/p>\n

\u2022 AdvSM (\u226520% incidence): edema, diarrhea, nausea, and fatigue\/asthenia. <\/span><\/p>\n

\u2022 ISM (\u226510% incidence): eye edema, dizziness, peripheral edema and flushing. <\/span><\/p>\n

 <\/span><\/p>\n

DRUG INTERACTIONS <\/strong><\/strong><\/span><\/p>\n

\u2022 Strong and Moderate CYP3A Inhibitors: Avoid coadministration of LuciAva with strong and moderate CYP3A inhibitors. If coadministration of LuciAva with a moderate inhibitor cannot be avoided, reduce dose of LuciAva in patients with GIST or AdvSM. <\/span><\/p>\n

\u2022 Strong and Moderate CYP3A Inducers: Avoid coadministration of LuciAva with strong and moderate CYP3A inducers.<\/span><\/p>\n

 <\/span><\/p>\n

USE IN SPECIFIC POPULATIONS <\/strong><\/strong><\/span><\/p>\n

Lactation: Advise not to breastfeed. <\/span><\/p>\n

 <\/span><\/p>\n

Storage <\/strong><\/strong><\/span><\/p>\n

Store at 20\u2103 to 25\u2103 (68\u2109 to 77\u2109), excursions permitted between 15\u2103 and 30\u2103 (59\u2109 and 86\u2109) [see USP Controlled Room Temperature]. Protect from moisture.<\/span><\/p>\n
